Vesicle-mediated protein trafficking is an essential process for distribution of newly synthesized proteins to various endomembrane compartments. In addition, it also plays a crucial role in reorganization of protein composition of these organelles. For this process, cargo proteins are delivered by vesicles. Many different types of vesicles are used in protein trafficking depending on the types of organelles. One of them is the clathrin-coated vesicles which are mainly used in trafficking among post-Golgi compartments. For delivery of cargoes by clathrin-coated vesicle, first cargoes have to be specifically sorted by cargo receptors and then clathrin has to be recruited to the donor compartments by adaptor proteins. Therefore, the cargo receptor and adaptor proteins are essential components of clathrin-mediated proteins trafficking at the post-Golgi compartments. In plants, vacuolar sorting receptors (VSRs) are essential in vacuolar trafficking of soluble cargoes from the TGN to the prevacuolar compartments. VSRs specifically sort vacuolar cargoes at the TGN. To recruit clathrin to the donor compartments, clathrin adaptor proteins are necessary. Many different types of clathrin adaptor proteins exist in plants. One of them is adaptor protein complexes (APs) composed of four different subunits. Another group of proteins is monomeric adaptors called Epsin-related proteins. They work at both the TGN and plasma membrane to recruit clathrin for vesicle formation. They can be divided into two groups, one for vacuolar trafficking working at the TGN and the other for endocytosis working at the plasma membrane. In the talk, I will present recent findings on the mechanism of AtVSRs and two types of adaptors proteins, APs and Epsin-related proteins in vacuolar trafficking and endocytosis in plants.
